Harvard Researchers Are Raising Questions About Why We Slow Down After 40
A molecule most people have never heard of may explain the energy crash so many of us accept as normal.
By the time most of us reach our forties, something has changed. The 3pm energy crash. The recovery that takes three days instead of one. The mental fog that wasn’t there a decade ago. Most of us are told, in one way or another, that this is simply what getting older looks like.
But a growing body of research out of Harvard Medical School suggests that explanation may be incomplete — and that the real cause is something far more specific than age itself.
The Molecule Behind the Crash
At the centre of this research is a molecule called NAD+ — short for nicotinamide adenine dinucleotide. It exists in every single cell in your body, and it plays a central role in how those cells produce energy.
The problem is that NAD+ levels don’t stay constant throughout life. They peak in your twenties, then begin a slow, steady decline — one that accelerates noticeably from your forties onward.
David Sinclair, a genetics professor at Harvard Medical School, has spent much of his career studying this exact decline. His research points to a repair enzyme called PARP-1 as a major driver — one that becomes more active in response to the stresses of modern life, and consumes NAD+ at an accelerated rate as it does.
Why This Is Only Now Getting Attention
NAD+ decline has been studied in longevity research for years, but it rarely reaches mainstream conversation. Part of the reason is that the science is genuinely complex. Part of it is that, until recently, there wasn’t a practical way to address it.
That is starting to change. NMN — nicotinamide mononucleotide — is a precursor molecule the body uses to rebuild NAD+. It has become one of the most closely studied compounds in the longevity field over the past several years.
But researchers have identified two problems with how most NMN products on the market are made. First, many use a fraction of the dose used in the studies showing real results — often 150 to 250 milligrams, compared with the 500 milligrams used in published research. Second, standard capsules are frequently broken down by stomach acid before the NMN ever reaches the bloodstream, meaning much of what people take may never actually be absorbed.
These two issues — dosage and absorption — are what a UK-based supplement called NadFuel was specifically formulated to address. It uses a full 500 milligram dose in a delayed release capsule designed to survive the stomach and reach the small intestine intact, where NMN absorption actually occurs.
